产品名称(CCK8)细胞增殖及细胞毒性检测试剂盒

应用详情

1、收集细胞，加细胞悬液100ul（约5000-10000 个细胞）到96 孔板（边缘孔用无菌水或PBS 填充）。每板设
对照（加100ul 培养基）。
2、置37℃，5% CO2 孵育过夜，倒置显微镜下观察。
3、每孔加入10 ul 待检测药物溶液， 37℃孵育。
4、每孔加入10 ul CCK-8 溶液，37℃孵育1-4 小时。
5、测定450 nm 各孔的吸光值，如无450nm 滤光片，可以使用450-490nm 的滤光片。
6、同时设置空白孔（培养基和CCK-8 溶液，无细胞），对照孔（不加药培养基和CCK-8 溶液，有细胞），每组
设定3-5 复孔。

产品描述

细胞增殖及细胞毒性检测试剂盒Cell Counting Kit-8 (CCK-8) 是应用新型的水溶性四唑盐2-(2-甲氧基-4-硝苯基)-3-(4-硝苯基)-5-(2,4-二磺基苯)-2H-四唑单钠盐快速高灵敏度检测细胞增殖和细胞毒性的比色检测产品。

本四唑盐在电子载体存在的情况下能够被线粒体内的一些脱氢酶还原成橙黄色的水溶性的Formazan，生成的Formazan量与活细胞数量成正比。细胞增殖越多越快，则颜色越深；细胞毒性越大，则颜色越浅。对于同样的细胞，颜色的深浅和细胞数量呈线性关系。

CCK-8溶液可以直接加入到细胞样品中，不需要预配各种成分，CCK-8溶液很稳定，对细胞没有毒性，可以长时间孵育细胞。CCK-8法测定细胞增殖或毒性试验中活细胞数目的灵敏度高，无放射性，检测细胞增殖实验的灵敏度比其它方法如MTT, XTT, MTS和WST-1高。
